a stream or a download counts as a download. a download is: one download, per item, per day, per IP address. if an item page has more than one file only the first file downloaded counts.

So, as I'm asking this because I upload my podcast on archive.org, If I have 45 downloads, I can say that it has been downloaded / played 45 times by different users?

not necessarily because the same user can do one download per day so the same person (or IP address) might have come back on different days and download.
